Meibomian glands (MGs), specialized sebaceous glands embedded in the eyelids, are an integral component of the ocular defense system, as their oily secretion product, meibum, is essential for protecting the eye surface. In spite of its importance for ocular health, the transcriptional changes that lead to meibum production by MG acinar cells (meibocytes) have not been characterized in a systematic way so far. By combining spatial transcriptomics (ST), pseudotime, and functional enrichment analysis, ...
The lipid-secreting sebaceous glands (SG) are indispensable for maintaining skin structure and function, and may influence whole body’s energy metabolism. SG deregulation is also associated with a number of skin disorders, including but not limited to acne. Here, we exploit the high cellular resolution provided by sebaceous hyperplasia to spatially characterize the human SG transcriptional program. By integrating single-cell spatial transcriptomics, pseudotime analysis, RNA velocity, and functional ...

BACKGROUND:Animal experiments and studies in alcohol dependent patients indicate that ghrelin signaling in the brain is causally involved in the regulation of alcohol reward and intake. Increasing ghrelin levels enhances alcohol craving and intake, blocking ghrelin receptors abolishes these effects. If ghrelin is also involved in non-dependent alcohol consumption in humans, though, remains unknown. The aim was therefore to investigate the relationship between ghrelin serum levels and alcohol ...

OBJECTIVES: Although patients with depression often suffer from sleep disturbances, most of them are not sleepy. Upregulation of brain arousal has been proposed as pathophysiological mechanism explaining sleep disturbances, inner tension, autonomic hyperarousal and anhedonia in depression. The aim of the current study was to examine the association between night-time sleep disturbances and brain arousal regulation the next day in depressed versus non-depressed subjects. METHODS: Twenty-eight ...

BACKGROUND: Reference intervals for leukocyte subsets from peripheral blood are helpful for the understanding of disease states and therapy effects. METHODS: We performed in-depth immunophenotyping for 608 healthy German adults from the Leipzig region from 40 to 79 years by 10-color flow cytometry (FCM) to gain reference information for various leukocyte subsets including subsets of granulocytes, monocytes and lymphocytes. RESULTS: First, we derived gender- and age-specific reference intervals ...
Recent technological advances in single-cell genomics make it possible to analyze cellular heterogeneity of tumor samples. Here, we applied single-cell RNA-seq to measure the transcriptomes of 307 single cells cultured from three biopsies of three different patients with a BRAF/NRAS wild type, BRAF mutant/NRAS wild type and BRAF wild type/NRAS mutant melanoma metastasis, respectively. Analysis based on self-organizing maps identified sub-populations defined by multiple gene expression modules ...
Single-cell transcriptomics has been used for analysis of heterogeneous populations of cells during developmental processes and for analysis of tumor cell heterogeneity. More recently, analysis of pseudotime (PT) dynamics of heterogeneous cell populations has been established as a powerful concept to study developmental processes. Here we perform PT analysis of 3 melanoma short-term cultures with different genetic backgrounds to study specific and concordant properties of PT dynamics of selected ...

Colorectal cancer (CRC) arising in Lynch syndrome (LS) comprises tumours with constitutional mutations in DNA mismatch repair genes. There is still a lack of whole-genome and transcriptome studies of LS-CRC to address questions about similarities and differences in mutation and gene expression characteristics between LS-CRC and sporadic CRC, about the molecular heterogeneity of LS-CRC, and about specific mechanisms of LS-CRC genesis linked to dysfunctional mismatch repair in LS colonic mucosa and ...

We present an extensive and detailed study on the gene expression landscape of peripheral blood of healthy individuals based on transcriptome data of 3,388 individuals screened in the population study LIFE between 2011 and 2014. For analysis we applied a neural network technique using self-organizing maps (SOM). Our home-made R-program ‘oposSOM’ enables to reduce the dimension of expression data from tens of thousands of genes to a few thousand ‘meta-genes’ and generates portraits of transcriptional ...

Gene expression of 270 patients with head and neck cancer Within the Leipzig Head and Neck Group (LHNG) we designed an observational cohort study to include consecutive patients with primary and metachronous secondary HNSCC of oral cavity, larynx, oro-, and hypopharynx. All tumors were pathohistologically classified as head and neck squamous cell carcinoma (ICDO-M-8070/3, 8071/3, 8083/3). Excluded from the study were patients with prior history of cancer other than HNSCC within the last 5 years. ...
